Fibre Type Specificity of Il-6 Gene Transcription during Muscle Contraction in Rat: Association with Calcineurin Activity
نویسندگان
چکیده
In this study, we quantified the transcription of the IL-6 gene in individual fibres and the associated changes in calcineurin activity assessed at cellular level during prolonged muscle contraction. Individual myofibres were isolated from plantaris and soleus muscles of rats at the end of an exhaustive running exercise (n=10), typed according to their myosin heavy chain isoform content, and compared to those of resting rats (n=10). Using real-time PCR analysis in individual fibres, a marked rise in IL-6 transcript levels occurred in type I and IIa fibres at the end of exercise (P<0.05). Transcription of the gene encoding for the modulatory calcineurin-interacting protein-1 (MCIP-1), a sensitive indicator of calcineurin activity, also mainly increased in type I and IIa fibres (P<0.05). Moreover, a slight increase in MCIP-1 mRNA levels was observed in type IIx (P<0.05). Fibre types determined by immunohistochemistry were qualitatively examined for glycogen content using Periodic Acid Shiff staining, and no direct relationship was found, at the cellular level, between glycogen content, fibre-type and IL-6 transcription. Our data clearly suggest that IL-6 gene transcription was mainly observed in early-recruited myofibres and that contraction-induced IL-6 transcription could be associated with enhanced calcineurin activity. ss a00 40 00 53 , v er si on 1 5 O ct 2 00 9

Recruitment determines the profile of fibre-type-specific genes expressed across the range of muscle fibres associated with slow, fast fatigue-resistant and fast fatiguable motor units. Downstream signalling pathways activated by neural signalling and mechanical load have been the focus of intensive research in past years. It is now known that Ca(2+)-dependent calcineurin-nuclear factor of acti...

Background: Myokines released from skeletal muscle have multiple metabolic and hypertrophic effects. On the other hand, one of proposed pathways for effects of exercise training on metabolic diseases is calcineurin signaling pathway. With considering to relation between interleukin-6 (IL-6) and calcineurin, the purpose of this study was to investigate whether the resistance training has an effe...
متن کاملCalcineurin activates interleukin-6 transcription in mouse skeletal muscle in vivo and in C2C12 myotubes in vitro.
Expression of the cytokine interleukin-6 (IL-6) by skeletal muscle is hugely increased in response to a single bout of endurance exercise, and this appears to be mediated by increases in intracellular calcium. We examined the effects of endurance exercise on IL-6 mRNA levels and promoter activity in skeletal muscle in vivo, and the role of the calcium-activated calcineurin signaling pathway on ...
متن کاملCalcineurin controls nerve activity-dependent specification of slow skeletal muscle fibers but not muscle growth.
Nerve activity can induce long-lasting, transcription-dependent changes in skeletal muscle fibers and thus affect muscle growth and fiber-type specificity. Calcineurin signaling has been implicated in the transcriptional regulation of slow muscle fiber genes in culture, but the functional role of calcineurin in vivo has not been unambiguously demonstrated. Here, we report that the up-regulation...
